Endotoxin is a specific contamination concern
Bacterial endotoxins are components associated with Gram-negative bacteria. They can provoke strong biological responses. An endotoxin test is designed to assess this particular category of contamination; it does not identify the peptide or establish its chemical purity.
“Pyrogen” is a broader term for substances capable of producing fever. Endotoxin is an important pyrogen, but not every pyrogen is endotoxin. A negative endotoxin result therefore cannot automatically be expanded into a claim that all pyrogenic substances are absent.
How is testing interpreted?
A laboratory selects an appropriate method and assesses whether the sample interferes with it. Results need meaningful units, a detection or quantitation capability and a justified acceptance limit. A number has little value if the report does not explain what the method could detect or whether it was suitable for that sample.
Examples include Limulus amebocyte lysate (LAL)-based approaches, such as gel-clot methods, and photometric or kinetic methods that assess a measurable assay response. The method must be suitable for the specific sample; a named assay alone does not establish that interference was excluded. FDA’s March 2026 guidance discusses these approaches and appropriate component and finished-product testing. Acceptance criteria depend on the product and regulatory context, rather than one universal limit for every peptide. FDA: Pyrogen and Endotoxins Testing, March 2026.

No. Sterility testing addresses viable microorganisms under the conditions of the test. Endotoxin testing addresses bacterial endotoxin. Killing bacteria does not necessarily remove endotoxin, and a low endotoxin measurement does not demonstrate the absence of viable organisms.
These tests also do not replace controls that prevent contamination. The quality of materials, water systems, manufacturing, packaging and handling can affect the final result. Testing is one part of a control strategy, not a way to convert uncontrolled manufacture into an assured process.
What should a certificate say?
It should identify the sample and method, state the result and units, explain the applicable criterion, and make clear any relevant limitations. “Endotoxin tested” without the result is less informative than a documented measurement. “Not detected” means not detected under the stated method and limit, not absolute zero.
No combination of endotoxin, sterility or chemical-purity results establishes clinical effectiveness. Nor does an endotoxin test authorize a research material for human administration.
Takeaway: Endotoxin testing answers a specific microbiological-quality question. It is distinct from sterility, identity, purity and clinical safety.
Review flag: Microbiology/analytical review required; no universal human-use threshold is supplied.
